Myeloproliferative diseases (MPDs) are a group of disorders characterized by cellular proliferation of one or more hematologic cell lines distinct from acute leukemia. Chronic MPD consist of 4 diseases, including polycythemia rubra vera (PRV), primary thrombocythemia (PT), agnogenic myeloid metaplasia (AMM), and chronic myelogenous leukemia (CML). Another name for AMM is myelofibrosis (MF). In some patients, conditions overlap and clear categorization may be difficult. MPD may evolve into one of the other myeloproliferative conditions and/or transform to acute leukemia. |

Phase I/II Study of Non-Myeloablative Allogeneic Hematopoietic Stem Cell Transplantation in Patients With Myelodysplastic Syndromes or Myeloproliferative Disorders |

This multicenter trial is evaluating the efficacy of non-myeloablative allogeneic hematopoietic stem cell transplantation in patients with myelodysplastic syndromes or myeloproliferative disorders. The incidences of graft-vs-host disease, graft rejection, and non-relapse mortality are also being studied. In addition, the efficacy of a strategy for donor lymphocyte infusion based primarily on defined criteria of persistent or progressive disease in these patients is being evaluated. |

Azacitidine Plus Phenylbutyrate in Treating Patients With Acute Myeloid Leukemia or Myelodysplastic Syndrome |

This phase I clinical trial is studying the safety and toxicity of azacitidine in combination with phenylbutyrate in patients with recurrent, refractory, or untreated acute myeloid leukemia or myelodysplastic syndrome. Researchers will also attempt to determine the minimal effective pharmacologic dose of azacitidine required to consistently inhibit DNA methyltransferase in this patient population. Preliminary clinical and/or laboratory data suggesting potential therapeutic activity of this combination regimen in these patients will be obtained. |

Bone Marrow Transplantation Plus Biological Therapy in Treating Patients With Chronic Myeloid Leukemia |

This phase II clinical trial is evaluating the effectiveness of bone marrow transplantation, chemotherapy, and biological therapy for chronic myeloid leukemia. The one-year event-free survival in patients with chronic phase chronic myeloid leukemia receiving sargramostim (GM-CSF)-treated autologous bone marrow transplantation, followed by GM-CSF and interferon alfa will be determined. Toxicity of this regimen will also be studied. |
